英语. The Agerolese cattle is a breed with an ancient origin and at the same time is an endangered breed, with only 121 cows in milking. The sister chromatid exchange (SCE) test has been used to detect chromosomal fragility in both humans and animals and to discover DNA damages caused by a variety of natural and artificial chemical compounds. Fifty Agerolese cattle (15 males and 35 females) from nine different farms of Naples province underwent to cytogenetic investigation by using the SCE test. Peripheral blood was cultured for about 72 h at 37.8 deg C in RPMI medium, enriched with FCS (10%), L-glutamine (1%) and concanavalin A (1.5%) as mitogen. BrdU (10 microng/ml) and colchicine were added 30 h before harvesting and slides were stained with acridine orange (0.1% in phosphate buffer, pH = 7.0). Metaphase plates were observed under a fluorescence microscope, captured with a digital camera and later processed. Upon 1,750 cells we studied, the total mean value of SCE/cell was 5.39 +- 2.58, a value which is much lower than the ones observed in other Italian cattle breeds and domestic species

Lo scambio intercromatidico (SCE) è stato utilizzato per valutare la fragilità cromosomica nell'uomo e negli animali e per individuale danni al DNA indotti da una varietà di composti chimici naturali e artificiali. Cinquanta bovini di razza Agerolese (15 maschi e 35 femmine) di nove allevamenti della provincia di Napoli sono stati sottoposti a un'indagine citogenetica mediante l'impiego del test SCE. Il sangue periferico è stato coltivato per circa 72 h a 37,8 gradi C in substrato RPMI, arricchito con FCS (10%), L-glutamina (1%) e concanavalina A (1,5%) come mitogeno. Alle colture sono state aggiunte la BrdU (10 microng/ml) e la colchicina 30 ore prima del termine e i vetrini sono stati colorati con arancio di acridina (0,1% in tampone fosfato, pH = 7,0). Le piastre metafasiche sono state osservate con un microscopio a fluorescenza, acquisite mediante una camera digitale e successivamente elaborate. Su 1.750 cellule studiate, il valore totale medio di SCE/cellula è risultato pari a 5,39 +- 2,58, molto inferiore a quelli osservati in altre razze bovine italiane e in altre specie domestiche.]
