Analysis of expressed sequence tags (ESTs) is an efficient approach for gene discovery, expression profiling, and development of resources useful for functional genomics studies. As part of the transcriptome analysis in olive flounder (Paralichthys olivaceus), we have conducted EST analysis using two cDNA libraries made from the spleen and head kidney. The assembly program ICAtools software was used to organize the redundant ESTs into overlapping contigs. The results showed that the 445 ESTs were composed of 50 clusters (spleen : 21, head kidney : 29) and 298 singletons (spleen : 174, head kidney : 124), suggesting that the overall redundancy of the library was 22%.
