Maceration of potato tuber tissue by Erwinia atroseptica mutants

Demonstration of Third Type Secretion System (TTSS) gene roles in maceration of potato tuber tissue, affected by Erwinia carotovora subspecies atroseptica is the aim of current work. This bacterium causes two economically important diseases of potato: blackleg (stem rot) and tuber soft rot. Recent discovery of special pathogens, and then TTSS, proteins and substrates of this system made more interests for studying their role in virulence of E. carotovora subsp.atroseptica. The responsible genes for this system play role in induction of hypersensitivity reaction and harpin secretion. There were used some E. carotovora subsp.atroseptica constructed mutant strains considering TTSS gene, cultured them in an appropriate media and temperature for overnight. After cultivation, bacteria were grown and transferred on potato slices. Infected slices incubated at appropriate temperature for overnight. In next step we measured and weighted maceration part of each slice for disrupted part and extracted liquids. Weighting disrupted tissues and measuring pectatelyase activity of them, finally using statistical methods we compared nominated strains together and to the original one. To summarize, hrpN, dspE, hrpj gene mutations increased bacterial ability for maceration of potato tuber tissue, while mutation remarks did not effect on bacterial pectolytic activity. It seems that disability of mutant strains in inducing of hypersensitivity response reaction is a reason for observed phenomenon
