Quantitative and qualitative analyses of the fractional proteins in different genotypes of sunflower (Helianthus annuus L.)

The aim of the investigation has been to study quantitative and qualitative composition of albumins, globulins and glutelins in photosynthetic and generative organs of different sunflower genotypes constituting the genetic system Cytoplasmic male sterility - pollen fertility restoration. Four genotypes were used: Three self-pollinated homozygous lines(SW501 - fertile line; SW501CMS - sterile line with cytoplasmic male sterility containing the mitochondrial gene orfH522; RW637Rf - line restoring pollen fertility, containing the nuclear gene Rf) and heterozygous F1 hybrids obtained from crossing SW501CMS and RW637Rf lines. Apical shoots at the stage of 2-4 leaves, photosynthetic organs (3-4 top leaves) and generative organs (sunflower heads) at the budding stage were analysed. Samples were taken at the end of 24 hours after exogenous treatment with 0.005 percent solution of gibberellic acid. The outcoming results allowed revealing that the content of protein fractions, differing in solubility, depends on genotype, plant organ and ontogenetic phase. Thus, the male sterile line is characterized by minimal content of all studied protein fractions in apex (ontogenetic phase of 2-4 leaves) as well as in leaves and heads (budding stage). The level of fractional proteins is higher in photosynthetic organs of all studied genotypes. The quantity of fractional proteins increases over the course of ontogenesis. The effect of the gibberellin is significant. It inhibits the proteosynthesis in RW637Rf line, whereas in all other genotypes it have a stimulating effect. Qualitative analysis of studied protein fractions showed that the product of the mitochondrial gene orfH522 is a polypeptide with relative molecular weight of 16 kDa, being a saltsoluble protein associated with cytoplasmic male sterility in sunflower and induced in the fertile lines by exogenous treatment with gibberellic acid.
