Detection of anthelmintic resistance for benzimidazoles in selected goat (Capra hiscus Linn.) farms in Cagayan Valley [Philippines] by microagar larval development test and allele specific-polymerase chain reaction

Anthelmintic resistance for benzimidazoles was detected in 31 selected commercial goat farms in Cagayan Valley using microagar larval development test (MALDT) and allele-specific polymerase chain reaction (AS-PCR). Sixteen farms were located in Isabela, 11 farms in Cagayan, and 4 farms in Nueva Viscaya. Species identification of gastrointestinal nematodes was performed using nested PCR and Hemonchus contortus came out to be the sole identified species of gastrointestinal nematode in all 3 provinces. Results of MALDT and AS-PCR benzimidazole resistance status in the 3 provinces indicated that there is no significant difference in proportion of detecting resistance per goat farm using both methods across 3 sites (Isabela, Cagayan and Nueva Vizcaya). In the present study, larger proportion of resistance was observed using MALDT, there were more farms that were detected as resistant compared to AS-PCR. Inability to detect higher resistance using AS-PCR might have been due to the few number of larvae (10 larvae per farm) used in the study. The most important recommendation in the study is to analyze more larvae from each selected farm to obtain a better picture of the benzimidazole resistance status of not only Haemonchus contortus but also other gastrointestinal nematodes like Trichostrongylus spp. in the different farms. Another equally essential recommendation is to clone and sequence the Beta tubulin gene of the H. contortus from the PCR product and compare it with the Beta tubulin sequence data in a foreign country.
