Efficient method for the simultaneous recovery of DNA and RNA from soil microbes in clayish and acid soils

Recently, molecular biology techniques have been widely used to study the structure diversity and the function of soil microbial communities. The basis of these techniques is the extraction of sufficient and high purity DNA or RNA from soil samples. Because of a high clay content and low pH in the soils of subtropical China, it is difficult to extract DNA or RNA from the soils from this region. In this study, by combining the pre-lysis washing procedure and the direct extraction method, DNA and RNA were simultaneously recovered from two typical soils in subtropical China, and the recovery efficiency was estimated. Analysis of the extracted DNA or RNA with enzyme digestion, polymerase chain reaction (PCR) and reverse transcription polymerase chain reaction (RT-PCR) showed that the purity of the nucleic acid was suitable for follow-up procedures. This provides a feasible way to further study microbial activities and function in the soils of subtropical China and similar elsewhere.
