Wild rice establishment technique for C4-ness mass screening

Healthy seedlings are essential for a successful screening for C4-ness traits in wild rice. The authors evaluated the effects of different potting media such as vermiculite, perlite, garden soil (dry), and puddled incubated garden soil on the growth of wild rice seedlings. Puddled soil incubated for 7d and watered with Yoshida culture solution was the best growing medium for wild rice seedlings. The method solved the iron deficiency problem in wild rice seedlings due to the uncontrolled pH encountered when using unpuddled soil or other medium that sits continuously in culture solution. The technique included dehulling and sterilizing seeds by dipping in a 1:1 mixture benomyl and mancozeb fungicides at the rate of 1 g product/ liter of water. The seeds were incubated at 30 deg C in the germination chamber for 1-2 d and sown in a 25.8 cm x 52.3 cm plastic tray (6 x 12 holes) filled with incubated puddled soil a day before seeding. To ensure vigorous seedling growth, the tray were immersed daily for 10 min in Yoshida culture solution and sprinkler irrigated with tap water as needed. The seedlings were grown up to the 5-6 leaf stage (2-3 wk) for evaluation for CO2 compensation point and vein density.
