Induction and germination of somatic embryos from zygotic and nucellar tissues of Helmar pummelo (Citrus maxima L.)

The Helmar pummelo of the Aklan State University is a newly registered pummelo variety by the National Seed Industry Council (NSIC). The variety has the following physical characteristics: fruit size - 106.03 mm; total soluble solid - 11.38 degree Brix; edible portion 57.66%, flesh color- white and flavor - sweet. In order to characterize the induction and germination process of the somatic embryos of the Helmar pummelo in vitro, an experiment was conducted using the Murashige and Skoog (MS) medium supplemented with 3 ppm BAP and 20 g/L sucrose. The study revealed that fully developed somatic embryos induced from the nucellar tissues yielded 3.14 shoots per culture and the zygotic tissues only developed 34.29% callus. The generants could be obtained from nucellar tissues of somatic embryos for the germination of shoots.
