Role of the S1-IAA9 transcription factor in fruit quality using reverse genetics in tomato

The present work is dedicated to the analysis of the transgenic plants of Micro Tom and Ailsa Craig in which an auxin transcription factor (Sl-IAA9) has been downregulated. Previous data had shown that the transgenic plants had entire leaves and parthenocarpic fruits. The fruits of the antisense lines of the Micro Tom culivar showed slower ripening rate and were smaller than the control. Biochemical analysis indicated that the antisense fruits had a higher sugar content, higher acidity and thicker pericarp. The ripe antisense fruits had a more intense colour than the wildtype. A panel test was performed and a significant difference was found concerning the texture: antisense fruits were catalogued as having a more floury texture. The same test revealed the fact that the antisense fruits were more aromatic. For the Ailsa Craig fruits a possible maturation delay was detected, the immature fruits being tagged as less aromatic compared to the fruits of the wild type cultivar at the same maturation level. This difference was no longer present for the ripe red fruits. In order to identify differentially expressed genes in tomato pericarp, we TOM1 oligo-slide microarrays containing 12.899 oligonuleotides were used. The results have shown that there are 440 differentially expressed genes; genes involved in maturation processes such as cell wall degradation, hormone regulation and other maturation related processes. Some of the genes that undergo changes in expression could be related to the phenotype.
