143 diploid and tetraploid genotypes of genus Solanum were characterised with the view of DNA polymorphism using for potential detection of potato somatic hybrids. Restriction analysis of CAPS marker SPUD237 resulted to detection of three variants of amplified nuclear DNA sequence polymorphism differentiate in presence and position of restriction site of AluI restrictase. Likewise, there is possible to differentiate the genotypes of Solanum bubocastanum from another species on the basis of SSCP analysis of rrn5/rrnl8-1 PCR product. RAPD analysis by means of OPN18 primer detected reliably interspecific and intraspecific polymorphism of DNA. Bulk segregation analysis differentiates the genotypes to three general clusters. The first cluster was generated only from all Solanum bulbocastanum genotypes. All these methods seem to be usable for potato somatic hybrids detection and they can be applicable only in concrete cases. Nevertheless, neither horizontal agarose electrophoresis nor SSCP analysis of intergenic spacer trnL/trnF detected polymorphism of PCR products and these results can predicate sequential conservativness of this spacer within genus Solanum. No somaclonal variability was detected in plants regenerated from protoplast culture originated from Solanum bulbocastanum 17.
