Nodal explants of Gardenia obtusifolia with single bud and at different order, the 3rd- 6th node from shoot tip, were cultured on MS medium supplemented with 0-20 micro mol/l BA. The results showed that both the order of nodes and BA concentrations had affected shoot multiplication. The 3rd and 4th node treated with 15 micro mol/l BA gave the highest shoot number of 3.6 and 3.0, respectively. Later, the new in vitro shoots were cultured on WPM in comparison with MS medium and resulted that the WPM provided better shoot multiplication. The effects of 0-10 micro mol/l BA in combination with 0-15 micro mol/l adenine sulfate (ADS) on shoot multiplication were conducted. It was found that 10 ?mol/l BA supplemented with 5 ?mol/l ADS gave the highest shoot number of 7.8 which was not significantly different with using 5 micro mol/l BA. These proliferated shoots could induce 3.8-6 roots on WPM supplemented with 5-15 micro mol/l IBA and gave 100 percent survival rate when transplanting to the greenhouse. For callus induction, young leaves were cultured on WPM medium supplemented with 0-20 micro mol/l NAA. The callus was sucessfully induced with 15-20 micro mol/l NAA under light condition and had better result than in the dark.
