Purpose of this research is to determine the optimal factors on rough rice bran cultivation i.e. pH, temperature and water content of Aspergillus japonicus SA22P3406. These factors were varied in the study of the optimization by response surface methodology followed by the central composite design. The optimum conditions of the pH, the temperature and the water content were 7.37, 36.0 deg C and 82.10 percent v/w of dry rough rice bran, respectively. These conditions were determined a sporulation curve. It was shown the fungal sporulation had less an average decadic logarithm of practical quantitative sporulation of 8.46 than with the estimated quantitative sporulation of 8.53 at the 5th day incubation.
