A field experiment based on the cotton-Bemisia tobacci interaction system was carried out in 2007 in Dunhuang City, Gansu Province, in order to study the physiological and biochemical characteristics of cotton(the cultivar ‘Dunmian V3') in chemical defense under the stress of the insect Bemisia tobacci. The dynamic changes of six physiological and biochemical indexes such as superoxide dismutase (SOD), and peroxidase (POD), and catalase (CAT), and ascorbate peroxidase (APX), and carotenoid (Car) in the cotton leaves in different population density were measured. The results showed that the activity of SOD, POD, CAT and APX in the cotton leaves, after influenced by the insect, were positively correlated to the difference of insect population density, however, carotenoid content was negatively correlated to the difference of insect population density. Such variations of SOD, POD, CAT, APX activity and carotenoid content of cotton leaves were more significant at the distance of 10 m than 20 m from insect sources.
