Gamma interferon (IFN-gamma) production protects cows infected with Neospora caninum against abortion. The present study analyzes interactions between IFN-gamma production and levels of plasma pregnancy-associated glycoproteins-1 (PAG-1) and IgG1 and IgG2 throughout gestation in Neospora-infected dairy cows. Plasma collected throughout gestation or until abortion was tested for antibodies against N. caninum, IgG1 and IgG2 isotypes, PAG-1 and IFN-gamma. GLM repeated measures analysis revealed that non-aborting Neosporaseronegative and -seropositive dairy cows (n=62) showing no IFN-gamma production exhibited higher (P=0.04) PAG-1 concentrations than seropositive cows producing IFN-gamma (n=14). Nonaborting seropositive cows producing IFN-gamma (n=10) showed higher (P=0.01) IgG2/IgG1 ratio than cows not producing IFN-gamma (n=11). A negative (P=0.027) effect of IFN-gamma production on IgG1 was also observed, while IFN-gamma production did not affect IgG2 levels. In contrast, higher levels of IgG2 than IgG1 antibodies were observed in aborting seropositive animals (n=10), both in those producing and not producing IFN-gamma. Our data indicate that in Neospora-infected dairy cows, the production of IFN-gamma correlates negatively and the production of antibodies is uncorrelated with plasma PAG-1 concentrations during gestation and that a Th1 cellular immune response, in which IgG2 antibodies prevail, could be protective against N. caninum abortion, but only in cows producing IFN-gamma.
