During cryopreservation spermatozoa suffer changes in their plasmatic membrane which cause a decrease in sperm quality and finally in their fertilizing ability. There are differences between species in their sperm cryoresistance. This is partly due to differences in plasma membrane composition, sperm from species cold-shock resistant present higher ratio cholesterol: phospholipids. In addition, after thawing sperm are in a state similar to capacitation, in which cholesterol is lost from the membrane. Increasing the cholesterol content of sperm plasma membrane could protect sperm during cryopreservation. The cholesterol can be added to plasmatic membrane by means of cholesterol-loaded cyclodextrin (CLC). With this technique, sperm cryosurvival increased in many domestic species, although results reported for boar sperm are contradictory. The objective in this study is to evaluate the motility during the cryopreservation process in boar sperm enriched in or deprived of cholesterol. The percentage of total motile spermatozoa (% TMS) and rapid progressive spermatozoa (% RPS) remained high until the equilibration with glycerol (87-93% TMS; 71-78% RPS) for all treatments. However, motility abruptly decreased after thawing (54-62% TMS; 50-56% RPS; P<0,0001), regardless of the treatment. In conclusion, neither cholesterol addition nor deprivation improves boar sperm quality after cryopreservation.
