Fibre plays an important role in the nutrition of the rabbit, not only as a source of nutrient but also as a regulator of the digestive tract, especially in the caecum where a wide variety of microorganisms are located. The aim of this study was to analyze the microbial diversity of caecal content and caecotrophes of growing rabbits fed on a diet with two levels of indigestible fibre (26 and 33%). Bacterial profile of ten animals was analyzed by DGGE and diversity was compared by the modified Shannon index. Caecotrophes were 71% similar to caecal content but diet modified this percentage, being the diet with the high level of FAD (33%) which showed the lowest percentage of similarity between them (66 %). However, nor diet neither source of the sample modified the biodiversity index (3.48 +/- 0.042 and 3.45 +/- 0.051 for caecal content and caecotrophes, respectively). Taking into account these considerations, soft faeces seem to be a good alternative to represent caecal content in studies of diet comparison.
