Effective Virus-free Seed Production by Meristem Culture at Clove Differentiation Stage of Garlic (Allium sativum L.)

This study was conducted to investigate the possibility on the seed garlic production by meristem culture in the clove differentiation stage of garlic plants. The traditional meristem culture is some problems such that it takes the high cost and labor input required for extraction of meristem, in vitro culture period and its difficult to bulblet formation. The clove differentiation stage of garlic plants is late-March to early-April in Southern regions in Korea. The clove weight and length of garlic plants in April 4 were 0.012 g and 4.84 mm, respectively. The meristem extraction time of this stage was cut down 47.8% as 2.4 hours/100 ea, compared with conventional method. Rate of in vitro bulblet formation was the highest as 150% at April 4 compared with others extraction time and number of bulblets over 0.4g was 16.0 ea/50 explants. Storage period of garlic plants for meristem extraction was possible for 120 days at 2±1℃. In the field cultivation of in vitro bulblets, growth and bulb production were no problems under different bulblet sizes. Bulblets over 0.6 g produced clove bulb over 78.0%. Thus, we are considerable that the meristem culture in the clove differentiation stage of garlic is very simple and effective for virus-free seed garlic production.
