Changes in seed quality and seed-borne pathogen of breeder seeds stored in cold room

The quality of breeder seeds (BS) is important in the successful production of foundation seeds (FS). Rigid supervision must be employed from production to post production operations to ensure high seed quality. Nineteen BS harvested in 2007 dry and wet season (DS and WS), dried at 12% moisture content (MC), then stored in cold room (14 to 16 deg C) were used in the study. Initial germination, MC determination, and blotter test were done prior to storage. Germination was done following the International Seed Testing procedure and fungal pathogen was evaluated using blotter tests with 400 and 200 seeds, respectively. Succeeding seed quality assessment was done every 3 months. BS produced in DS showed higher initial germination than those produced in WS. The higher germination noted was also observed even after 3, 6, and 9 months of storage. Average germination rates in BS harvested in DS and WS after 9 months were 96 and 91%, respectively. Thus, low temperature storage maintained germination above the 85% standard. Incidence of fungal observed was higher in BS harvested in WS than DS. Regardless of season, percentage occurrence of fungal pathogen decreased as storage duration was extended. Likewise, Aspergillus flavus, a storage fungus was observed after 3 months with minimal occurrence. The level did not increase probably owing to the low MC of the BS.
