Determination of vegatative compatibility groups and patoypes of Verticillium dahliae isolates | Zeytinden elde edilen Verticillium dahliae izolatlarının vejetatif uyum grupları ve patotiplerinin belirlenmesi

英语. Verticillium wilt, caused by Verticillium dahliae, is the most serious disease in olive cultivation areas in Turkey. Workers using isolates from different hosts described characterisation by vegetative compatibility (VCG) groups, which complement traditional pathogenicity tests. VCG refers to the ability of individual fungal strains to undergo mutual hyphal anastomosis and form viable heterokaryons. In this study two hundred and eight isolates of V. dahliae from olive trees were taken for vegetative compatibility analysis using nitrate non-utilizing (nit) mutants. One isolate did not produce a nit mutant. Nit mutants of 219 isolates were tested against tester strains of internationally known vegetative compatibility groups (VCGs)1A, 2A, 2B, 3, 4A and 4B, and also paired in many combinations among themselves. One hundred and nintyfive of the isolates (89.45%) were strongly compatible with T9, the tester strain of VCG1A, and thus were assigned to VCG1A. Ten isolates were assigned to VCG2A and seven isolates to VCG4B. One isolate was heterokaryon self-incompatible (HSI) and five isolates could not be grouped to any of the VCGs tested. Pathogenicity tests were realized on Manzanilla, V.dahliae susceptible cultivar. Isolates of VCG1 group,including T9 tester isolate, showed high rate of disease (3.12-98,43% ) and defoliating symptoms at the inoculation tests made in climatic chamber.

土耳其. Verticillium dahliae (Kleb.)zeytin alanlarında görülen en önemli hastalıklardan biridir. Araştırıcılar çeşitli konukçulardan izolatları, patojenisite testlerinin tamamlayıcısı olan vejetatif uyum (VCG) grupları ile karakterize etmiştir. VCG, fungal streynlerin hiflerinin birleşebilme ve canlı heterokaryonlar oluşturma kabiliyetini ifade eder. Bu projede çoğunluğu Batı Anadalu Bölgesinden olmak üzere 219 V.dahliae izolatının genetik farklılığı nitrat kullanmayan (nit) mutanlar üzerinden değerlendirilmiştir. Sadece bir izolat nit mutantı üretmemiştir. Bu yüzden geriye kalan 218 adet V. dahliae izolatının nit mutantları ise uluslar arası tanınan VCGs 1A, 2A, 2B, 3, 4A ve 4B tester streynleri ile ve pek çok kombinasyonda kendi aralarında testlenmiştir. 195 adet izolat (%89.45) VCG1A'nın tester streyni T9 ile kuvvetli uyum VCG1A grubuna ait bulunmuştur. On izolat VCG2A'ya ve yedi izolat ise VCG4B'ye ait bulunmuştur. Bir adet izolat heterokaryon self-incompatible (HSI) olmuş ve 5 adet zeytin izolatı ise testlenen herhangi bir VCG'ye ait olmamıştır.
