Effect of Orchiectomie on histomorphology of the kidney | Ratlarda Kastrasyonun böbrek histomorfolojisine etkisi

英语. The aim of the present study was to determine the histological and morphological differences in kidney tissue of young adult rats, which went through an orchiectomie operation. A total of 36 male Sprague-Dawley rats were used and rats were divided into 4 subgroups, two control groups (n=16) and two experimental groups (n=20). Whereas rats in the experimental groups went through the orchiectomie operation, rats in control groups did not have an operation and were kept under the same conditions with the experimental groups. Kidney tissue samples were taken from rats of both the control groups and experiment groups, one and two months after the orchiectomie operation. In order to determine histometric changes, Periodic Acid Schiff (PAS) staining was performed. Light microscope (Leica DMLB) equipped with image analyzing system (Leica Q Win Standart) was used to measure various morphometrical parameters (the numbers of renal corpuscles and tubuli per unit area, the diameter of renal corpuscles, proximal tubule, henles loop and collecting tubule) in order to observe the changes in the kidney histology. Intensity of PAS-positive granules within the cytoplasm of cells constituting the proximal tubule was found to be higher in the experimental groups than that determined for the control groups. Morphometric measurements taken 1 month after then the orchiectomie operation indicated that while the numbers of renal corpuscles per unit area was higher in the experimental groups than that of the control groups, the diameter of the renal corpuscles, henles loop and collecting tubule was lower when compared to the control groups. Morphometric measurements taken 2 months after then the orchiectomie operation indicated that whereas there is an increase in the diameter of the renal corpuscles and collecting tubule, diameter of proximal tubule and henles loop decreases when compared to the control groups. In conclusion, these results demonstrate histologic and morphometric changes in the kidney tiussue as a consequence of orchiectomie operation.

土耳其. Araştırmada, genç erişkin dönemde kastrasyon yapılan ratların böbrek dokusunda meydana gelen histolojik ve morfometrik farklılıkların incelenmesi amaçlandı. Materyal olarak 2 aylık 36 adet erkek Sprague-Dawley rat kullanıldı. Ratlar 2 kontrol (n=16) ve 2 deney (n=20) olmak üzere toplam 4 guruba ayrıldı. Deney gurubundaki ratlara kastrasyon işlemi yapıldı. Kontrol gurubundaki ratlara ise herhangi bir işlem yapılmadan eşit ortam koşullarında tutuldu. Kastrasyondan 1 ile 2 ay sonraki dönemlerde hem kontrol hem de deney guruplarından doku örnekleri alındı. Histometrik değişimleri belirlemek amacıyla Periodic Acid Schiff (PAS) boyama metodu uygulandı. Işık mikroskobu (Leica DMLB) ve buna bağlı Görüntü analiz sistemi (Leica Q Win Standart) ile morfometrik ölçümler yapılarak (birim alanda korpuskulum renis ve tubulus sayıları ile korpuskulum renis, proksimal tubul, henle kulpu ve toplayıcı borucuk çapları) böbrek histolojisinde meydana gelen değişimler incelendi. Yapılan çalışmada, kastrasyondan hem 1 ay hem de 2 ay sonraki dönemlerde, proksimal tubulleri oluşturan hücrelerin sitoplazmalarında kontrol gruplarına göre PAS pozitif sitoplazmik granüllerin çok yoğun olduğu gözlendi. Morfometrik ölçümler sonucu ise, kastrasyondan 1 ay sonra alınan örneklerde birim alandaki korpuskulum renis sayısı kontrollere göre artarken, korpuskulum renis, henle kulpu ve toplayıcı borucuk çaplarının azaldığı saptandı. Kastrasyondan 2 ay sonra yapılan ölçümlerde, korpuskulum renis ve toplayıcı borucuk çapları artarken, tubulus proksimalis ve henle kulpu çaplarının azaldığı görüldü. Sonuç olarak, kastrasyonun böbrek histokimyası ve morfometrisine yönelik oluşturduğu değişimler ortaya konmuştur.
