The application of in vitro mutagenesis technique by gamma irradiation for dianthus breeding

To create new varieties of dianthus for commercial flower production purpose, callus of dianthus varieties SP25 (Dianthus caryophyllus Make up), SP2 (Dianthus caryophyllus Carmel), SP11 (Dianthus caryophyllus Niva) were cultured in medium of shoot multiplication and treated with five different doses of gamma rays (0,5, 1, 2, 3, 4 krad). After five subculture regenerations (M1V5), the plantlets have been transfered to the rooted media. The evaluating of irradiated explants in in vitro and in the field showed tht: In in vitro, gamma rays was affected to growth and height of shoots as its multiplicative coefficient, especially 4 krad. Besides gamma rays also has been delayed and decreased rooting induction of shoots. In the field, after 7 weeks, the irradiating was affected to plant height. In two SP2, SP25 varieties with 3 and 4 krad doses of gamma rays have appeared solid mutants about shape of stens and leaves. All of these mutants will be the rich materials to establish new varieties of dianthus in the near future.
