The selection of high molecular weight glutenin Dy-subunits for hexaploid wheat (Triticum aestivum L.) by DNA marker

Variation in high molecular weight glutenin subunits in hexaploid wheat (Triticum aestivum L.) is extensive, mainly at the locus Glu–1B. On the contrary variation at loci Glu–1A and Glu–1D is low although proteins encoded by these loci also very intensively affect parameters of technological quality of wheat grain. Therefore programme for new HMW–GS and relevant alleles started and within it discovered new HMW–GS 1Dy12.3 in old wheat cultivar Noe. Subsequently difference in nucleotide sequence between new allele and alleles 1Dy12 and 1Dy10 were identified. PCR primers were designed on base of high homology of HMW–GS DNA sequences 1Dy10, 1Dy12, and 1Dy12.3, respectively. Sequence within coding region of Dy subunits revealed differences in the central domain where the 1Dy12.3 differs from 1Dy10 and 1D12 subunits by deletion of coding di- or hexa-peptid, respectively.
