Bovine Leukemia Virus (BLV) is a retrovirus that belongs to the Retroviridae family and cause lymphoma and most frequently diagnosed malignant neoplastic disorder in dairy cattle. The infection, therefore, is economically important because of the loss of export markets that require BLV-free cattle. Several studies have been carried out in an attempt to determine whether BLV causes neoplastic diseases in humans, especially through the consumption of milk from infected cows. Establishment of simple available and valid diagnostic test is a tool for achieving a successful BLV control program strategies. In this study, Immunoperoxidase staining method has been established for detection of antibody in bovine serum. BLV-infected FLK (Fetal Bovine Kidney) cells were cultured in 96 well mircoplate and they were considered as the source of virus and antigen. To block endogenous peroxidase activity, the cells were immersed in diluted blocking solution. Diluted serum samples were treated with BLV coated antigen after that with Rabbit _Anti Cow HRP Conjugate secondary antibody. The incidence of antibodies to bovine leukemia virus in sera samples of cattle from Isfahan province was determined using the Indirect Immunoperoxidase staining method. Of 405 sera samples, 309 (76.3%) were positive. Since ELISA verification's current kit was used in this study and samples have also been examined and agreement between two tests was found 87.2% .The sensitivity and specificity was shown 88.8% and 79.7%, respectively. The hypothesis of sensitivity and specificity 90% and 85% respectively has been conformed.
