Analysis of Genimic Structure of HA98 gene and evaluation of Intron sequences of Hyalomma anatolicum anatolicum.

Previous studies indicated that Bm86 protein of Boophilus Microplus tick was as immunogene in domestic animals. Hyaloma anatolicum anatolicum, the main vector tick, is the most dominant tick species in Iran and has a wide geographic distribution in this country. Extensive studies have been done to identify immunogenic proteins of ticks such as Hyaloma. One of them is Bm86-like protein (HA98) of Hyaloma anatolicum anatolicum. The sequence of the cDNA of this gene has been determined, but there is no data about this gene. The aim of this study was to investigate the HA98 gene structure in the chromosome and genome level. Based on available sequences, several primers designed in different part of the sequences for amplification and isolation the full-length (with potential interons) of HA98 gene. DNA was extracted from tick egg, amplified by PCR, cloned and sequenced. The results indicated that no intros presented in HA98 gene (unlike most eukaryotic genes) and contained at least one ORF that code for one protein with 664 amino acids.
