In vitro propagation of Opuntia ficus indica

Opuntia is a valuable type of forage which grows in arid and semiarid regions during periods of drought and shortage of herbaceous plants. Cactus is rich in carbohydrates and calcium and cactus is also used to combat desertification. The aim of this work was micropropagation of Tuna (Opuntia ficus _ indica) to provide mass production of Tuna. For this reason, explants dissected from young cladodes were cultured on medium and results showed that explants had satisfactory establishment with 90%. Different combinations of shoot proliferation were tested for shoot development and elongation and our results showed that the best proliferation coefficient was 6 in this medium. Rooting was achieved in Basal medium (1-2 cm length) whereas satisfactory rooting (8-10 cm length) was obtained in rooting medium. The highest percentage of rooting (100%), number of roots/explant (4), maximum total length of root/explant (10 cm) was obtained in rooting medium. At acclimatization period, survival rate of the ex-vitro plantlets rooted in different rooting media was 100%. Plants were successfully established in soil and acclimatization was accomplished under greenhouse condition.
