Gel filtration, electrophoresis, and amino acid analysis of acid-soluble and acid-insoluble fractions of rice endosperm glutelins.

Rice endosperm glutelins were investigated by gel filtration, electrophoresis, and amino acid analysis. Salt-soluble and alcohol-soluble proteins were removed by percolating mixture of rice flour, celite and fine sand with 5% sodium chloride solution and 60% ethanol on Maes' column. Glutelins were extracted with 4% lactic acid and 0.1% potassium hydroxide. About half amount of glutelins were soluble in lactic acid, and another half were insoluble in acid but soluble in alkali. Gel filtrations of whole glutelins gave four peaks; II and III were derived from acid-soluble, and I and IV were from acid-insoluble fractions. Two fractions have quite different patterns in amino acid composition. Acid-soluble fractions were rich in aspartic acid, leucine, and arginine, while acid-insoluble fractions were rich in lysine, threonine, alanine, and tyrosine. Acid-insoluble fractions must be nutritionally superior to acid-soluble fractions. Appreciable differences among varieties were detected in GF patterns, electrophoresis, and amino acid composition.
