Viability studies on seeds and embryonic axes of Lansium domesticum Corr.

Studies were carried out on seeds of three varieties of Lansium domesticum namely duku, duku langsat and langsat. Seeds were desiccated at room temperature (27 degree plus minus 2 degree C) for 0, 8, 16, 24, 32, 40 and 48 hours. After each desiccation period, moisture content was determined and viability was assessed. It was observed that duku seeds survived to 38 percent moisture content (15.6 percent germination) whereas duku langsat seeds rapidly lost viability below 44 percent moisture, while those of langsat survived desiccation to 26 percent moisture (46.7 percentage germination). Non-cryopreserved axes desiccated to less than 38 percentage moisture content did not show any viability. Four cryopreservation methods viz. desiccation, encapsulation-dehydration, vitrification and slow-freezing were studied using langsat embryonic axes. No axes survived cryopreservation in any of the four methods employed.
