In tthe present work a series of parameters were studied, in order to determine a method to multiply explants of pink shallot (Allium ascalonicum L.), which were cultivated in vitro, allowing the development of an abundant proliferation of new shoots. Three groups of experiments were done. The purpose of the first one was to disinfect the plant vegetal material: One group of treatments consisted on the use of Zonalin (1,5 ml/l), Agallol (5 g/l), and Bacilan (7 g/l), for 10 or 15 min. Finally, Bacilan and Zonalin were combined together with three doses of calcium hypochlorite (0.0; 0.7 or 1.4%) for 15 minutes. In the second group of experiments, two media were tested: B5 (Gramborg et al, 1969) and 0,5MS (Murashige and Skoog, 1962). In the third group of assays, the effect of phytohormones upon shoot proliferation was tested. The response of the shallot explants was tested with B5 medium was supplemented with NAA (0.0; 0.1; 0.2 and 0.4 mg/l) and BAP (0.0; 1.0; 2.0 and 4.0 mg/l). Afterwards, 0.5 MS medium was supplemented with two concentrations of BAP and 2iP (0.25 and 0.5 mg/l). Finally, the effect of supplementing 0.5MS and B5 media with BAP (0.0; 1.0; 2.0 mg/l) was studied. Results proved that 5% of sodium hypochlorite used for 15 minutes, allowed the total disinfection of explants, and the normal development of shoots and roots, which did not occur with the other disinfectants. B5 medium was better than 0.5MS, increasing the number of shoots per culture. The addition of NAA (0.2 mg/l), together with BAP (4.0 mg/l) to the B5 medium, produced the highest rate of shoot multiplication (5.4 shoot per explant). The higher shoot proliferation occurred when 2iP was used. Nevertheless shoot proliferation was not different for the two concentrations used.
