Tissue culture of Citrullus lanatus (Thunb.) Matsum and Nakai var. Charleston grey (watermelon).

Very young fruit, cut 3-5 mm in length was observed to be the best explant for the tissue culture of watermelon var. Charleston Grey, as compared with other plant parts such as : leaf, stem, shoot apex, floral bud, pedicel and ovary. The best culture medium is Revised Murashige and Skoog (RMS) supplemented with 2.5 kinetin (ki) ppm, 0.25 IBA ppm in pH 5.7 + under direct light laboratory condition. The lower endocarp with rind of very young fruits when used as explants produced plantlets with: Yellowish-green stem, shoot differentiation and pufuse creamy-white to white root-like structures in RMS medium initially supplemented with 2.5 ppm Ki plus 10.00 ppm NAA after five subcultures in RMS medium with 2.5 ppm Ki. The plantlets were twice subcultured in the same medium plus 0.25 ppm NAA to produce luxuriant growth of plantlets with roots. For the induction of more roots each plantlet was subcultured in RMS supplemented NAA ppm and 0.25 with 2.5 ppm Ki plus 0.25 ppm NAA and 0.25 ppm IBA.
