Effect of phospholipase A2 supplementation on penetration of zona-free hamster eggs by bovine sperms.

The supplementation of BO medium with phospholipase A2 of disparate sources and its subsequent effect on penetration of zona-free hamster eggs by bovine sperms was investigated. In the first experiment using ejaculated spermatozoa from 2 bulls and an enzyme derived from snake (Naja naja) venom, the penetration rate was increased, though it was not significant (17.8 and 26.1% without enzyme; 24.1 and 30.5 with 10 mU/ml of enzyme; 18.0 and 32.2% with 100mU/ml of enzyme in bull A and B, respectively). In the second experiment using frozen cauda epididymal spermatozoa from 2 bulls and Phospholipase A2 of disparate sources, the penetration rate was increased in medium supplemented with 10 mU/ml of enzyme derived from snake venom (2.5 and 6.7% without enzyme; 7.9 and 36.1% in bull C and D, respectively) and porcine pancreas (2.8 and 19.8% in bull C and D, respectively). The increase observed in bull D was significant (p<0.01). On the other hand, 10mU/ml of the enzyme derived from bee venom had no significant effect on the penetration rate (3.1 and 12.6% in bull C and bull D, respectively). These results do suggest that phospholipase A2 supplementation of culture media does facilitate sperm penetration of zona-free hamster eggs.
