Proteinuria in healthy cattle: a comparative study with two different reagent strips

It has been stated that normal urine in healthy bovine contains only small amounts of protein that clinical standard test could not detect. It also has been cited that alkaline urine could cause false positive results for urine protein. We have studied the urine protein excretion in 127 clinically healthy Holstein cows during the first month of lactation. The animals included in the study were at different lactation (from first to fifth lactation) and were maintained in intensive milk production and fed by uni-feed. The animals had normal dry matter intake, and parturition had been developed without problems. Urine samples were obtained by catheterism. Semiquantitative urine protein were analyzed immediately by two different reagent strips and then stored in plastic tubes at -20 deg C until colorimetric analyses for protein. The mean urine protein was 30.31 +/- 22.44 mg/dl. The lactation number has some effect on urine protein, being higher the mean value in the first lactation animals and lower in the fifth lactation animals, in fact there was statistically significant differences between them. Comparing the results obtained by reagent strip and conventional colorimetry there was a low correlation, both in identifying the negative values as in the discrimination of different existing concentrations. The sensibility and specificity to identify proteinuric animals was not too high for both reagent strips, with a high percentage of false positives. In conclusion, the studied reagent strips are not very useful to analyze proteinuria in the bovine.
