Protein analysis of stem bark from three citrus varieties infected with Citrus tristeza virus in Pakistan

Plant virus synthesizes the structural and functional protein during infection process. Therefore a molecular study was carried out to analyze the effect of citrus tristeza infection on host protein. Citrus samples infected with Citrus tristeza virus (CTV) (Genus Closterovirus) of three citrus varieties viz., Kinnow mandarin (Citrus raticuIata Blanco), sweet orange (C. sinensis L.) and grape fruit (C. paradise Macf.) from Punjab and Khyber Pakhtunkhwa in Pakistan were collected and confirmed through ELISA. Bark samples were prepared and analyzed through SDS-PAGE for alteration in host protein. There was no significant or little alteration in the bark protein of the (TV-infected and healthy samples from citrus trees. The bands produced on gel electrophoresis showed a slight alteration for the peptide mapping of sweet orange from Punjab and Khyber Pakhtunkhwa infected with CTV. Bands in these samples showed their tendency towards higher molecular weight. Overall the bands were approximately in the range of 15 kDa. It can be concluded that CTV has not been characterized on strains basis in Pakistan yet, therefore slight alteration in host protein of infected samples may reveal the presence of mild strain of this virus.
