[Biocatalytic method of producing natural food dyes from vegetable raw materials] | Биокаталитический способ получения натуральных пищевых красителей из растительного сырья

Studied was a possibility to produce natural food dyes from sweet sorghum stems and palm fruits fruits by using enzyme preparation of microbial origin: CK and BC based on cellulose, VIS based on pectinesterase and UF based on pectintranseliminase. For the greatest yield of anthocyans from sweet sorghum the biocatalysis length is 2 h for BC, UF and VIS and 1.5 h for CK. The optimal temperature of fermentolysis is found to be 55 degrees C for BC and 50 degrees C for remaining enzyme preparations. In case of extracting carotinoids from palm fruits the water duty varied from 1:5 to 1.25. For all the studied enzymes the optimal ratio of raw materials:water is 1:10. The greatest yield of carotinoids (92% of the original content) was achieved by using VIS. The optimal doses of enzymes (% to the raw material weight) are VIS – 0.2; BC – 0.15; UF – 0.05 and CK – 0.1. The most rational length of fermentolysis is 2 h for all enzymes. The substrate composition of raw materials explains the difference in the effect of enzymes: prevailing cellulose in sweet sorghum and insoluble pectin in palm fruits. Extracts of anthocyans from sweet sorghum have been obtained by the method of biocatalysis having the maximum yield by using CK (89%). The optimal process parameters: water duty is 1:20, t is 50 degrees C, dosage is 0.1%, and the length of fermentolysis is 1.5 h. VIS appeared to be the most effective for extracting carotinoids from palm fruits (yield – 90%). The rational regime: water duty is 1:10, t is 50 degrees C, dosage is 0.2%, and the length of fermentolysis is 2 h. during treating sweet sorghum the presence of water for action of hydrolases is sufficient, during treating palm fruits it is necessary to use a 15% ethanol solution at the stage of fermentation and 50% solution at the stage of extracting. The best result for red food dye samples was achieved by using CK for treating sweet sorghum (38.41 CU), in case of palm fruit samples the best orange color saturation was achieved by using VIS (51.26 CU).

Исследовали возможность производства натуральных пищевых красителей (ПК) из стеблей сорго сахарного (СС) и плодов пальмы масличной (ПП) с применение ферментных препаратов (ФП) микробного происхождения: ЦК и БЦ на основе целлюлазы, ВИС на основе пектинэстеразы и УФ на основе пектинтрансэлиминазы. Для наибольшего выхода антоцианов из СС продолжительность биокатализа составляет 2 ч для БЦ, УФ и ВИС и 1,5 ч для ЦК. Оптимальная температура ферментолиза установлена 55 град. C для БЦ и 50 град. C для остальных ФП. В случае экстракции каротиноидов из ПП варьировали показатель гидромодуля от 1:5 до 1:25. Для всех исследуемых ФП оптимальное соотношение сырье : вода равно 1:10. Наибольший выход каротиноидов (92% от исходного содержания) получен при использовании ВИС. Оптимальные дозы ФП (% к массе сырья) составляют: ВИС – 0,2; БЦ – 0,15; УФ – 0,05 и ЦК – 0,1. Наиболее рациональная продолжительность ферментолиза – 2 ч для всех ФП. Различие в действии ФП объясняется субстратным составом сырья: преобладанием целлюлозы в СС и нерастворимого пектина в ПП. Методом биокатализа получены экстракты антоцианов из СС с максимальным выходом при использовании ЦК (89%). Оптимальные параметры процесса: гидромодуль - 1:20, t - 50 град. C, доза - 0,1%, продолжительность ферментолиза - 1,5 ч. При выделении каротиноидов из ПП наиболее эффективен оказался ВИС (выход – 90%). Рациональный режим: гидромодуль – 1:10, t – 50 град. C, доза – 0,2%, продолжительность – 2 ч. При обработке СС достаточно присутствия воды для действия гидролаз, при обработке ПП необходимо использовать 15%-ный р-р этанола на стадии ферментации и 50%-ный – на стадии экстракции. Лучший результат для образцов ПК красного цвета получен при использовании ЦК для обработки СС (38,41 усл.ед.), в случае образцов ПП лучшая цветовая насыщенность ПК оранжевого цвета получена при использовании ВИС (51,26 усл.ед.).
