Standard karyotype and DNA markers for sex determination of tilapia fish (Oreochromis aureus)

Tilapia (Oreohcromis aureus) is an economic freshwater fish. The sex this kind of fish could not be discriminated at hatchery until fry stage. This research initiated genetic techniques for suitable sex discrimination in both male and female of O. aureus. Somatic karyotypes of both sexes were the same and they are asymmetrical with 2n=44. They composed of 1 pair of acrocentric macrochromosome and 21 pairs of acrocentric microchromosomes. Nucleolar organizer regions were on the short arm of a microchromosome pair. No sex chromosomes were observed. This discovery is different from the previous report which classified sex chromosome system of this fish as ZZ/ZW. Four DNA markers for sex discrimination were established using SSH (suppression subtractive hybridization). They were PCR (polymerase chain reaction) based marker and could verify sex of Tilapia at the fry stage.
