Isolation and characterization of Solanum lycopersicum leafy cotyledon1-like gene and its relationship with germination

The tomato (Solanum lycopersicum) is the most commonly commercially produced vegetable all over the world. However, it has many production limitations; biotic and abiotic limitations in fruit seed and other developmental stages. Physiological disorders come, as serious limitations, in advanced rank among other constraints. One of these physiological disorders is seeds sprouting during fruit formation, resulting in huge economical losses for the producers. Therefore, the studying of the regulators of this physiological phenomenon is a very important target and tool. It is well documented, in Arabidopsis thaliana and some other species, that the Leafy cotyledon (LECs) gene group plays key regulation roles during seed development stages. However, nobody mentions the exact functions of this group in tomato seed development and/or seed germination stages. Moreover, there is strong evidence for AtLEC1 regulation of fatty acids biosynthesis. Studying LEC1-LIKE (LEC1L) in tomato will open the door for the production of improved tomato functional food. This is the motivation behind this study. In this study, the full-length coding sequence of tomato LEC1L was isolated and cloned. A wide range comparative analysis of DNA and predicted protein sequences has been done to finally prove that we have cloned a SlLEC1L which is an ortholog of AtLEC1L and other LEC1L genes. Furthermore, in silicon structural, functional, protein-protein interaction and protein-chemical interaction predictions have given ideas about the functional and conserved domains of SlLEC1L which is working as a CCAAT box binding transcription factor for many CCAAT box promoter dependent genes.
