Evaluation of immunization of K99 antigen, toxin and whole killed E. coli K99 antigen Diarrhea caused by Escherichia coli K99 in calves especially in the first days after birth, is important, due to resulting from casualties and economic losses. In this study, after isolating strains, with strains submissions from Razi Institute attempte to purify the K99 fimbrea antigen. Results showed 16 (7.5%) of 280 Escherichia coli isolated had a K99 fimbrea. With the same number of valence single antiserum O20 and O8 agglutination test was negative. But antiserum O101 and O 9 showed a positive reaction. Purification results with different methods of heating, Bead, sonication and heating was found 60 degree of centigrade in 20 minutes was achieved purify antigen than others methods.To prepare purified antigen, chromatography was used was very fruitful. Thus an affinity chromatography column with sepharose 4 B gel activated by cyanogen bromide only one band to the K 99antigen was obtained. An antigen level was obtained between 1.5 to 2.3 mg per liter variable. Bands specified in each method using SDS-Page was visible. Although many studies related to infection and virulence of this bacterium has been done, but except in some cases prevalence, already in the field of common O sero-group in Iran much study has not been done. With purified antigen K 99 also found no case in Iran. Considering that this antigen could provide background for the vaccine to provide, hope in the near future it will investigate.
