Endophytic actinobacteria of Cellulomonas genus in an artificially created association with meristem potato | Эндофитные актинобактерии рода Cellulomonas в искусственно созданной ассоциации с меристемным картофелем

Studied was an ability of the auxins producing strain Cellulomonas sp. 7, isolated from the inner tissues of roots of winter rye (Secale cereale L.) on sod-podzolic soil and manifests growth-stimulating activity in the host plant, to interact with other crop - potato (Solanum tuberosum L.), propagated in vitro. The study was conducted on two varieties of potato - Rozhdestvensky and Darik. Creating of microbe-plant association in vitro was carried out by coating of cell suspension of studied isolates on the surface of the explant. As a result of inoculation Cellulomonas sp. 7 actively colonizes the surface and internal tissues of the shoots (0.5x10E9 and 4.2x10E6 CFU/g, respectively on 30th day after treatment) and roots (3.4x10E7 and 9.4x10E3 CFU/g) of plant of Rozhdestvensky variety. Population density of colonization of the phylloplane (2.6x10E9 CFU/g) and rhizoplane (0.7x10E7 CFU/g) of potato Darik by cellulomanades was on the same level as potato Rozhdestvensky; however in the internal tissues of the shoots of this variety bacteria were found in smaller number (2.1x10E3 CFU/g) than that of the Rozhdestvensky variety and in surface-sterilized roots did not show up at all. The number of cellulomanades in phytosphere of variety Rozhdestvensky remained at a high level throughout the experiment. Stable colonization by studied isolate of phytosphere of potato Darik observed for only 2 passages. So, demonstrated was the ability of Cellulomonas sp. 7 to interact with potato meristem in influence its growth in vitro. Treatment of explants Cellulomonas sp. 7 promotes an increase in shoots height, the mass and the coefficient of plant propagation. Significant growth-promoting effect of the inoculation of plants is achieved by selection of the optimal concentration of inoculum. The density of population of cellulomanades of plant tissues and the stability of the associated as created is greatly variety specific.

Исследовали способность продуцирующего ауксины штамма 7 Cellulomonas sp., выделенного из внутренних тканей корней озимой ржи (Secale cereale L.) на дерново-подзолистой почве и проявлявшего в отношении растения-хозяина ростстимулирующую активность, вступать во взаимодействие с картофелем (Solanum tuberosum L.), размножаемым in vitro. Исследование проводили на сортах картофеля Рождественский и Дарик. Создание микробно-растительной ассоциации in vitro осуществляли путем нанесения на поверхность эксплантов суспензии клеток изучаемого изолята. Cellulomonas sp. 7 активно колонизировал поверхность и внутренние ткани побегов (0,5x10E9 и 4,2x10E6 КОЕ/г соответственно; на 30 сутки с момента обработки) и корней (3,4x10E7 и 9,4x10E3 КОЕ/г) растений сорта Рождественский. Плотность заселения целлюломонадами филлопланы (2,6x10E9 КОЕ/г) и ризопланы (0,7x10E7 КОЕ/г) картофеля Дарик была на том же уровне, что и картофеля Рождественский, однако, во внутренних тканях побегов этого сорта бактерии обнаруживались в меньшем, чем у сорта Рождественский, количестве (2,1x10E3 КОЕ/г), а в поверхностно стерилизованных корнях не обнаруживались совсем. Численность целлюломонад в фитосфере сорта Рождественский сохранялась на достаточно высоком уровне в течение всего эксперимента. Стабильная колонизация изучаемым изолятом фитосферы картофеля Дарик наблюдалась на протяжении только 2 пассажей. Итак, показана способность Cellulomonas sp. 7 вступать во взаимодействие с меристемным картофелем и влиять на его рост in vitro. Обработка эксплантов Cellulomonas sp. 7 способствовала увеличению высоты побегов, массы и коэффициента размножения растений. Существенный фитостимулирующий эффект от инокуляции достигается путем подбора оптимальной концентрации инокулюма. Плотность заселения целлюломонадами растительных тканей и стабильность созданной ассоциации в значительной степени сортоспецифичны.
