Development of full-length infectious clone of Iranian isolate of chicken anaemia virus.

Chicken anemia virus (CAV) is the causative agent of a disease characterized by aplastic anemia and generalized lymphoid atrophy with concomitant immunosuppression and frequently is complicated by secondary viral, bacterial, or fungal infections. Economic losses due to CAV-induced disease are associated with increased mortality, the cost of antimicrobial therapy to control secondary infections and poor growth. In this project for the first time in Iran, CAV was isolated in MSB1 cell line, and then the viral genome cloned into the bacterial vector pTZ57R. The DNA sequence of the insert was determined and analyzed. The clone was tested in the cell culture and in a day-old chick trial to confirm its infectious nature. The result revealed the isolated CAV strain is closely related to a strain from Bangladesh and has not vaccine origin. The poor growth of the isolated stain in cell culture is likely associated with of QQ HV region of the viral VP1 gene. The electroporation of the clone caused CPE in MSB1 cells and its injection into day-old chicks had manifestations specific to CAV infection. This is the first report of the development of an infectious clone of an animal virus in Iran, which is a fundamental step for the development of a mutant vaccine strain or a DNA.
