The probiotic strain Lactobacillus reuteri: selection of the technological regimes of cultivation | Пробиотический штамм Lactobacillus reuteri: подбор технологических режимов культивирования

Выделен пробиотический штамм Lactobacillus reutеri, относящийся к одному из немногих видов аутохтонных лактобацилл, присущих человеку, и перспективный в производстве кисломолочных продуктов с пробиотическими свойствами. Разработана питательная среда для его культивирования (гидролизованное молоко, полученное путем гидролиза ферментом Alcalase; инулин; дрожжевой экстракт), позволяющая получать наибольшее количество жизнеспособных клеток - 9,11 lg КОЕ/куб.см. Изучена динамика размножения Lactobacillus reutеri в ферментере при различных значениях рН (5,6; 5,8; 6,0; 6,2; 6,4; 6,6) и температурах (32, 35, 37, 39, 41 град. C); определены оптимальные условия, обеспечивающие интенсивное накопление жизнеспособных и активных клеток. Инокулят готовили на среде MRS и вносили в количестве 5% во всех опытах. Количество клеток Lactobacillus reutеri определяли на среде ГМК-1 в анаэробных условиях при 37 град. C в течение 3-5 дней. Наибольшее количество клеток Lactobacillus reutеri было достигнуто при культивировании штамма при рН 5,8-6,2: при поддержании рН 5,8 количество клеток составило 9,18 lg КОЕ/куб. см, при рН 6,0 - 9,24 lg КОЕ/куб. см, а при рН 6,2 - 9,15 lg КОЕ/куб.см. Изменение температуры существенно воздействовало на процесс размножения клеток. При температуре 35 град. C наибольшее содержание клеток составило 9,14 lg КОЕ/куб. см, а при 37 град. C - 9,18 lg КОЕ/куб. см. Более высокие (39 и 41 град. C) или низкие (32 град. C) температуры культивирования негативно сказывались на накоплении клеток в питательной среде. Интервал температур 35-37 град. C и активной кислотности 5,8-6,2 оптимальны для культивирования Lactobacillus reutеri и обеспечивают высокий прирост клеток.

Probiotic strain Lactobacillus reuteri, relating to one of a few species of autohtotic lactobacillus typical for human and prospective in the production of fermented milk products with probiotic properties was isolated. The nutrient medium for its cultivation was developed (hydrolyzed milk obtained by hydrolysis with the enzyme Alcalase, inulin, yeast extract) that allows producing the largest quantity of the living cells - 9.11 lg CFU/cub. cm. Dynamics of Lactobacillus reuteri propagation in the fermenter at different pH (5.6, 5.8, 6.9, 6.2, 6.4, 6.6) and temperatures (32, 35, 37, 39, 41 deg. C) was studied and optimal conditions providing intensive accumulation of living and active cells were determined. The inoculate was prepared on the MRS medium and in all experiments it was introduced at the amount of 5 %. The number of Lactobacillus reuteri cells was determined on the GMK -1 medium in anaerobic conditions at 37deg. C within 3-5 days. The largest number of Lactobacillus reuteri cells was achieved by the strain cultivation at pH 5.8-6.2: if maintaining the pH at the level of 5.8 the number of cells was 9.18 lg CFU/cub. cm but at pH 6.2 it was 9.15 lg cfu/cm3. Changes of the temperature considerably effected the process of cell proliferation. The largest cells number - 9.14 lg CFU/ cub. cm was at the temperature 35 deg.C and at 37 deg. C the cells number was at the level of 9.18 lg CFU /cub.cm. The higher cultivation temperatures (39 and 41deg. C) or the low temperatures (32 deg. C) negatively affected cells accumulation in the medium. The temperature range of 35-37 deg.C and active acidity of 5.8-6.2 are considered to be optimal for cultivation of the Lactobacillus reuteri and provide high cells growth.
