Идентификация сортов сельскохозяйственных культур по электрофоретическим спектрам запасных белков | Identification of crop varieties by the electrophoretic spectra of reserve proteins

Работы по использованию запасных белков зерна как маркеров в изучении генетических ресурсов растений, идентификации сортов пшеницы и других злаков были начаты во Всесоюзном НИИ растениеводства под руководством В.Г. Конарева в 1967 г., когда в институте была создана лаборатория белка и нуклеиновых кислот, переименованная позже в отдел молекулярной биологии. Преимущество белка как генетического маркера связано с тем, что белок является первичным и непосредственным продуктом генетической системы. Для идентификации сортов пшеницы, ячменя, овса и других зерновых культур используют электрофорез в вертикальных пластинах 6,5% ПААГ в кислой среде. В статье обсуждаются некоторые итоги и перспективы использования электрофореза запасных белков семян злаков – проламинов – в идентификации сортов, а также пути стандартизации этого метода. На примере пшеницы и ячменя показана перспективность использования его в селекции, семеноводстве, семенном контроле, а также в генных банках для изучения, систематизации и документации генофонда культурных растений и их диких родичей, для контроля за генетической стабильностью образцов, выявления дублетов или генетически близких образцов. К настоящему времени в виде белковых формул зарегистрировано более 5000 образцов пшеницы, ячменя, овса из коллекции ВИР; составлено 20 каталогов белковых формул, в том числе 7 в электронном виде. Отмечена роль академика РАСХН В.Г.Конарева (1915-2006) и руководимого им коллектива в разработке отечественных и международных (в сотрудничестве с ISTA – International Seed Testing Association) стандартных лабораторных методов семенного контроля (идентификации сортов), основанных на электрофорезе запасных белков семян.

英语. The work on the use of reserve protein of grains as markers in studies of plant genetic resources and identification of wheat varieties and other cereals were initiated at N. I. Vavilov All-Russia Institute of Plant Genetic Resources (VIR) under the leadership of V. G. Konarev in 1967, when the laboratory of protein and nucleic acids was established in the Institute, later to be renamed the Department of molecular biology. The advantage of the protein as a genetic marker is due to the fact that protein is the primary and immediate product of the genetic system. To identify varieties of wheat, barley, oats, and other crops electrophoresis in vertical plates of 6.5% PAAG in an acidic environment was used. Some results and prospects of the use of electrophoresis of reserve proteins of seeds of cereals (prolamines) in the identification of the varieties and also ways of standardizing this method are discussed. For example, wheat and barley are the prospects of its use in breeding, seed production, and seed control, as well as in gene banks for study, systematization, and documentation of the gene pool of cultivated plants and their wild relatives, to monitor the genetic stability of samples, detection of doublets or genetically close samples. To date, in the form of protein formulas registered more than 5,000 samples of wheat, barley, and oat from VIR collection; composed of 20 directories, protein formulas, including 7 in electronic form. The role of the academician of Russian Academy of Agrarian Sciences V. G. Konarev (1915-2006) and led team in the development of domestic and international (in collaboration with ISTA – International Seed Testing Association) standard laboratory methods of seed testing (identification of varieties), based on the electrophoresis of reserve proteins of seeds is noted.
