Diagnostics of trichinosis in the early stages of larval development | Диагностика трихинеллеза на ранних стадиях развития личинок

俄语. Материалом для исследования служили образцы мышечной ткани крыс, экспериментально зараженных Trichinella spiralis. Всего трихинеллами в дозе 10 л/г было заражено 18 беспородных белых крыс массой 90-100 г. Для проведения ранней диагностики при трихинеллезе крыс убивали с 6 по 24 день после заражения. Компрессорным методом исследовали наиболее поражаемые и доступные мышцы: диафрагму, массетер и грудные мышцы при увеличении (х 25, 50, 100). На каждый срок сделаны микрофотографии личинок на срезах. Затем образцы мышц задних конечностей массой 50 г исследовали методом автоматизированного пептолиза (АП) на аппарате Гастрос. По окончании цикла работы аппарата при микроскопировании учитывали количество выделенных личинок и их морфологическое развитие. Выделенными личинками 16-18-дневного возраста были заражены мыши (биопроба). Через 35-36 сут тушка мыши полностью подвергалась АП для выявления трихинелл. При АП мышечной ткани крыс, инвазированных личинками T.spiralis, установили, что личинки в возрасте с 10 по 15 сут после заражения данным методом не выявляются. Единичные трихинеллы, начиная с возраста 16 дн.. выявляются только при микроскопировании. Массовое выделение трихинелл начинается с 17-го дня после заражения, когда большая часть личинок достигает инвазионности. Неинкапсулированные трихинеллы в возрасте 16 дн. становятся инвазионными, что показало успешное заражение белых мышей личинками, выделенными после АП. По результатам биопробы основная масса личинок становится инвазионными к 19-20 дням после заражения. Таким образом, методом АП мышечной ткани легко диагностировать заражение животных неинкапсулированными личинками трихинелл, что затруднительно или невозможно при компрессорном исследовании. Применение АП наиболее эффективно при достижении личинками инвазионности.

英语. Muscle tissue samples from rats experimentally infected with T. spiralis served as a test material. Altogether, 18 outbred white rats weighing 90-100 g were infested at the dose of 10 l/g. To make the early diagnosis of trichinellosis, rats were killed on the 6th – 24th day of infestation. The compression method was used to study at magnification (х 25, 50, 100) the most infected and accessible muscles: diaphragm, masseter and pectoral muscles. Photomicrographs of larvae sections were taken for each term. Then, muscle samples from the hind legs weighing 50 g were investigated by the automatic peptolysis (AP) method using the apparatus Gastros. After the end of operation cycle, the number of released larvae and their morphological development were microscopically detected. Mice were infected with released larvae aged 16 to 18 days (bioassay). 35-36 days later, the mouse corpse was examined with the use of AP to determine trichinas. The AP of rat muscle tissue infected with larvae of T. spiralis shows that larvae of 10 to 15 days of age after infestation are not detected. Single trichinas from the age of 16days, can be only microscopically detected. Massive release of trichinas begins since the 17th day after the infestation when most larvae reach the invasion stage. Non-encapsulated trichinas aged 16 days become invasive; this was proven by the successful experimental infestation of white mice with larvae released by the AP. According to the results of bioassay, most larvae become invasive on the 19-20th day of infestation. Therefore, the AP of muscle tissue is used to diagnose easily the animal infestation with non-encapsulated larvae of T. spiralis, which is rather difficult and impossible by using the compression method. The use of AP is the most effective for the period when larvae reach their invasion stage.
