Genetic polymorphism and apple mosaic virus prevalence in the gardens of Belarus

Genetic polymorphism and apple mosaic virus prevalence in the gardens of BelarusApple trees of different cultivars growing in Belarus were tested using RT-PCR for Apple mosaic virus, an important economically and common pathogens in commercial orchards. Viral infected apple trees (1.5 %) were detected in the modern horticultural plantations and were not found among the old trees aged more than 50 years. The fragments of ApMV coat protein gene were cloned and sequenced. The analysis of their nucleotide sequences showed that the identity between them ranged from 98.9 to 99.6 %. The analysis of evolutionary relationships of nucleotide sequences obtained in this study and their homologs from the GenBank database revealed that the similarity between the viral genotypes was observed both between the isolates from various plants growing in the same geographical region, as well as between the isolates obtained from the same plant species, growing in different geographic regions. This picture was probably formed as a result of two methods of apple mosaic virus propagation: vegetative and mechanical.

俄语. С помощью метода ОТ-ПЦР обнаружено, что среди деревьев из действующих промышленных и маточно-черенковых садов 1,5 % заражены вирусом мозаики яблони. У деревьев старше 50 лет инфекция не выявлена. Степень идентичности вирусов мозаики яблони, распространенных на территории Беларуси, определенная с помощью сиквенирования фрагментов гена, кодирующего белок оболочки, колебалась в пределах от 98,9 до 99,6 %. Анализ эволюционных взаимоотношений полученных нами нуклеотидных последовательностей и их гомологов из базы данных GenBank показал, что сходство между генотипами вируса наблюдается как у изолятов, выделенных из разных растений, растущих в одном географическом регионе, так и у изолятов, выделенных из одного вида растений, растущих в различных географических регионах. Это может быть следствием двух способов распространения вируса мозаики яблони – вегетативного и механического.
