The objective of the research was to conduct phage monitoring of cheese-making enterprises with the purpose of studying the parameters of interaction of phage associations (PA) adequately reflecting the phage pool of the enterprise and lactic acid bacteria. The research objects were cheese whey substrates, positioned as multistrain bacteriophage substrates (MBS). The results of the study of phage content of MBS selected in two cheese-making enterprises of the Altai region are presented. The presence and titer of phages in the substrates were determined by conventional methods of phagolysis using liquid and agar-nutrient media. 142 collection (indicator) cultures of mesophilic lactococci in relation to the PA, presumably contained in MBS, were screened. The high contaminating phage background of MBS and considerable differentiation of lytic parameters (spectrum/level) of PA at each enterprise were determined. Indices of lytic activity (ILA) of PA S1 24/04/13/v5, S2 24/04/13/v7 and S4 27/05/13 defined in relation to the Lc. lactis (73 strains) were 0.26, 0.23 and 0.19 respectively. The same index for three PA in relation to Lc. diacetylactis (50 strains) was lower and equaled 0.04. The values of ILA in PA B2 13/05/14; B4 15/07/14 and B6 16/09/14 for total sampling (128 strains) were in the range from 0.06 to 0.28. Almost the same variability of ILA corresponded to the sampling of Lc. lactis (76 strains). The variation range of ILA of the same PA for 52 cultures of Lc. diacetylactis was from 0.04 to 0.35. The obtained data confirmed the uniqueness of phage pools at various enterprises, high probability of cell phagolisis of starter associations of microorganisms and the necessity to improve methods of protection against phages in cheese-making.

В задачи работы входило проведение фагомониторинга сыродельных предприятий с целью изучения параметров взаимодействия фаговых ассоциаций (ФА), адекватно отражающих фаговый пул предприятия, и молочнокислых бактерий. Объектами служили субстраты подсырной сыворотки, позиционируемые как многоштаммовые фагосодержащие субстраты (МФС). Приведены результаты изучения фагосодержания МФС, отобранных на двух сыродельных предприятиях Алтайского края. Наличие и титр фагов в субстратах определяли общепринятыми методами по факту фаголизиса на жидких и агаризованных питательных средах. Проведен скрининг 142 коллекционных (индикаторных) культур мезофильных лактококков по отношению к ФА, предположительно содержащимся в МФС. Выявлен высокий контаминирующий фаговый фон МФС и значительная дифференциация литических параметров (спектра/уровня) ФА каждого предприятия. Индексы литической активности (ИЛА) ФА С1 24/04/13/в5; ФА С2 24/04/13/в7 и ФА С4 27/05/13, определенные по отношению к выборке Lc. lactis (73 штамма), составили соответственно 0,26; 0.23 и 0,19. Этот же показатель для данных 3 ФА относительно выборки Lc. diacetylactis (50 штаммов) был ниже и составил 0,04. Значения ИЛА в ряду ФА Б2 13/05/14; ФА Б4 15/07/14 и ФА Б6 16/09/14 для суммарной выборки (128 штаммов) лежали в диапазоне от 0,06 до 0,28. Практически такая же вариабельность ИЛА соответствовала и выборке Lc. lactis (76 штаммов). Диапазон варьирования ИЛА этих же ФА по отношению к 52 культурам Lc. diacetylactis составил от 0,04 до 0,35. Полученные данные подтвердили уникальность фаговых пулов различных предприятий, наличие высокой вероятности фаголизиса клеток заквасочных ассоциаций микроорганизмов и необходимость совершенствования методов защиты от фагов в сыроделии.
