Association of betaine aldehyde dehydrogenase 2 with accumulation of 2-acetyl-1- pyrroline and other aromatic compounds in rice (Oryza sativa L.)

Gene-specific markers coupled with robust phenotyping techniques deliver an ideal combination in characterizing grain quality traits in rice such as aroma. Using two allele-specific markers for BADPH2, two sets of recombinant inbred lines (RILs) were genotyped. Out of the 158 RILs derived from two aromatic rices Kai Noi Leuang (KNL) and Hom Nang Nuane (HNN), 66 had the 8-bp deletion at the 7th exon, 88 possess the 806-bp deletion across the 4th and 5th exons of BADH2 while four were heterozygous for both mutations. Using the badh2.11marker, predicted segregation ratio (aromatic:non-aromatic, 1:3) was observed in the FS RILs of KNL and the non-aromatic Thasano 1 (TSN 1). Using GAS Chromatography. Mass Spectrometry (GC-MS), the volatile compound 2-acetyl-1-pyrroline (2 AP) was quantified in the F7 RILs and found to be normally distributed and significantly different among haplotypes. Grain dimensions were distinct between KNL and HNN and normally distributed across the F7 RILs but were not significantly different between the F7 RILs with badh 2.1 and badh 2.11 genotypes. Using the two-dimensional GC coupled with time-of-flight-MS with Head Space Solid-phase Microextraction sampling, discriminative metabolites were detected in KNL, HNN and the F7 RILs suggesting a difference in volatile profiles of aromatic varieties with different BADH2 alleles.
