Development of the method of the indicator phages using for the identification of Salmonella spp. | Разработка способа использования индикаторных фагов для идентификации сальмонелл

英语. Research on the development of a phage identification scheme for Salmonella spp. was carried out by the Perm Veterinary Diagnostic Center in the period from 2014 to 2017. There were used strains of Salmonella spp., Escherichia coli, Shigella flexneri, Proteus spp., Staphylococcus aureus and Listeria monocytogenes (total of 131 strains). For the preparation of bacteriophages, white laboratory mice weighing 16-18 g (n = 22) were used. For purposes of investigation, an initial bacterial suspension containing 10E8 cells/cm3 was prepared by the Densi-La-Meter (Erba Lachema, Czech Republic) from the daily agar cultures of the microorganisms studied. Next, a series of successive dilutions was made to obtain a slurry containing 10E1 cells/cm3. As a result, an inoculum containing 10 cells of Salmonella was added to 9.0 cm3 of selenite broth and a similar volume of RVS- broth, an indicator phage in an amount of 0.1 to 3.0 cm3 was added. As a result, it was established that the combined incubation of Salmonella and the eponymous phage for 24 + 3 h in selenite and RVS-brothes promotes the lysis of Salmonella spp., which can serve as a diagnostic sign for the identification of Salmonella. Having conducted a similar experiment with microbial associates, visualized were varying degrees of turbidity and / or the presence of sediment, which led to the conclusion that the proposed method for identifying the desired bacteria is highly sensitive. The developed method for identification of Salmonella with the use of isolated bacteriophages surpassed the classical analogue for a number of indicators: sensitivity, efficiency, material and labor costs, and allowed saving 24 h of time.

俄语. Исследование по разработке схемы фагоидентификации бактерий рода Salmonella проводилось в Пермском ветеринарном диагностическом центре в 2014-2017 гг. Работали с референтными и полевыми штаммами Salmonella spp., Escherichia coli, Shigella flexneri, Proteus spp., Staphilococcus aureus и Listeria monocytogenes (всего 131 штамм). Для получения бактериофагов использовали белых лабораторных разнополых мышей массой 16-18 г (n=22). В целях исследования из суточных агаровых культур изучаемых микроорганизмов с помощью оптического прибора Densi-La-Meter (Erba Lachema, Чехия) готовили исходную бактериальную суспензию, содержащую 10E8 МТ/см3. Далее делали ряд последовательных разведений до получения суспензии c 10 МТ/см3. В итоге инокулюм, содержащий 10E1 МТ сальмонелл, вносили в 9,0 см3 селенитового бульона и аналогичный объем RVS-бульона, добавляли индикаторный фаг в количестве от 0,1 до 3,0 см3. В результате установили, что совместная инкубация сальмонелл и одноименного фага в течение 24 +-3 ч в селенитовом и RVS-бульонах способствует лизису бактерий р. Salmonella, что может служить диагностическим признаком идентификации сальмонелл. Проведя аналогичный опыт с микробами-ассоциантами, визуализировали разной степени помутнение и/или наличие осадка, что позволило сделать вывод о высокой чувствительности предложенного способа идентификации искомых бактерий. Разработанный способ идентификации сальмонелл с использованием выделенных бактериофагов превзошел классический аналог по ряду показателей: по чувствительности, эффективности, материальным, трудовым затратам и позволил сэкономить 24 ч времени.
