Development of sweet potato (Ipomea batatas (L.) Lam.) genotypes with high storage root anthocyanin content through marker-assisted selection

Five sweet potatoes genotypes (1 traditional and 4 advanced lines) were intercrossed in a reciprocal manner and the offspring of each were evaluated for storage root in a reciprocal manner and the offspring of each were evaluated for storage root anthocyanin accumulation through gene-based markers and chemical analysis and root and yield through phenotyping. Dendrogram of sweet potato genotypes showed high dissimilarity between 'Marinas' (P1) which is traditional and the other four parental which are advanced lines. Al tested anthocyanin-related markers used were detected among purple fleshed parental(P2, P3, and P5), however, the markers failed to accurately determine purple fleshed progenies. In addition, the gene-based markers were closely observed among genotypes with pigmented shoot parts. Row evaluation in two seasons have identified progenies processing both high yield and stability, namely Gen. 19 and Gen. 50. Ranking of genotypes considering the two traits using Base index generated five promising progenies which are Gen. 23, 65, 50,, 62, and 22. Negative association between the two traits of interest was observed. Hence, increasing the anthocyanin concentration in sweet potato flesh through breeding have unfavorable effect on yield. It is highly recommended to conduct a replicated and multi locational trial of these genotypes for both qualitative and quantitative traits Marker-assisted selection using gene-based markers should be done through RT-PCR on plant tissues of interest.
