Agrobacterium-mediated transformation of nipah virus surface glycoprotein in tobacco (Nicotiana tabacum)

Nipah virus (NiV) is a paramyxovirus, capable of causing considerable morbidity and mortality in a number of mammalian species, including human. The surface glycoprotein (G) of Nipah virus plays a key role in viral entry and induction of neutralization antibody in mammalian hosts. In this study, the Agrobacteriummediated transformation of Nipah virus surface glycoprotein (G) into tobacco was carried out. A virus G-gene cassette was constructe,. the G gene was driven by CaMV35S promoter and a Nos polyA terminator at the 3" end. Tobacco (Nicothiana tabacum) transformation was carried out by co-cultivating the tobacco leaf discs with transformed Agrobacterium tumefaciens strain LBA4404. Tranformants selection was done in MS medium containing 100g/ml kanamycin. Transformants were found proliferating and developing shoots in MS medium supplemented with 0.1mg/L NAA and 1 mg/L BAP. PCR screening showed the majority of the regenerated shoot containing the transformed Nipah virus G-gene.
