Phytate did not affect fecal endogenous excretion of zinc but suppressed its true absorption in rats

We detennined the effect of dietary phytate on true absorption and fecal endogenous excretion ofzinc in rats using a stable isotope ofzinc (67Zn). Rats were given diets mainly consisting of casein and starch. Sodium phytate was added at 1.2 % into the diet. Rats were surgically implanted an osmotic pump to infuse 67Zn for 1 week at a rate of 24 Ilg/day. Feces and urine were collected during infusion to detennine total zinc by atomic absorption spectrometer and 67Zn enrichment by an inductively coupled plasma mass spectrometer (ICP-MS). Since fecal and urinary enrichments of 67Zn became stable 4 days after the initiation of infusion, fecal endogenous excretion and true absorption of zinc were calculated from total zinc and 67Zn enrichment in fecal and urinary samples collected between 5 and 7 day of infusion. Dietary phytate did not affect fecal endogenous zinc but suppressed true absorption of zinc. These results indicated that dietary phytate decreased the apparent absorption of zinc through suppressing its true absorption.
