2003

Various factors affecting gene transformation through hypocotyl explants of Citrus grandis were investigated to optimize gene transformation. Those factors included strains of Agrobacterium, bacterial densities, tranformation enhancer concentrations and co-culture conditions. The result showed that hypocotyl explants infected with Agrobacterium strain EHA 105 demonstrated higher level of transient GUS expression than those infected with AGLI and LBA 4404 and strain EHA 105 was used in all subsequent experiment as the basis for identifying the most appropriate condition for transformation. Higher percentage of GUS expression was obtained with explants inoculted with Agrobacterium EHA 105 (OD600 0.2) for 40min. The best co-culture condition was the incubation of the explants for three days in culture medium supplemented with 200 uM acetosyringone.
