Effects of N6-Benzyladenine (BA) on in vitro culture of Spilanthes acmella

The effects of N6-benzyladenine (BA) on multiple shoot formation of Spilanthes acmella were studied. Axillary buds of the plant were used as explants which were surface sterilised using mercuric chloride (HgC12,0.08% w/v) followed by 15% Clorox for 5 and 15 minutes respectively. The aseptic explants were cultured on Murashige and Skoog (MS) medium supplemented with BA (0.0, 2.0, 4.0, 6.0, 8.0 and 10.0 mg/l) for induction of multiple shoots over a period of five weeks. Multiple shoots were formed in these media supplemented with 2.0, 4.0, 6.0 and 8.0 mg/l of BA. Results indicated that the addition of 6 mg/l of BA or more caused retardation and swelling of explants which eventually resulted in abnormal structures and death. The number of shoots formed after sub-culturing in the fifth week was found to be higher in those explants undergoing shoot separation as compared to non-separated individual shoots. MS medium supplemented with BA of 2 mg/l or less was the most suitable for the production of multiple shoots of S. acmella.
